Concentration, cellular exposure and specificity of organelle selective fluorescent dyes in fungal cell biology

چکیده

This technical focus article discusses the importance of concentration, cellular exposure and specificity for application organelle selective fluorescent dyes in fungi using DNA, membrane cell wall stains as examples. Nonetheless, presented considerations are generally applicable to all applied living cells. The association a dye with its target molecule impairs consequently function. Effective time key factors that influence biocompatibility any dye. Prominent molecules frequently used staining targets fungal biology are: (i) DNA nuclear labelling, (ii) α-/β-glucans chitin (iii) phospholipids plasma endomembrane labelling. In combination live-cell imaging settings reduce light stress, i.e. excitation intensities times set minimum still achieves good signal-to-noise ratios, is low dosage markers so called “vital dyes” essential visualising processes an artefact-free fashion.
